The study tested an early-intervention combo for Alzheimer’s disease in a triple-transgenic (3xTg-AD) mouse model: focused ultrasound to temporarily open the blood–brain barrier (FUS-BBBO) plus delivery of a small amyloid-binding compound called Re-1, targeting the hippocampus over 10 biweekly sessions across 5 months. In this work, the combination group showed encouraging trends in behavior, suggesting potential cognitive benefits even when changes in amyloid and tau pathology weren’t consistently significant across groups. Interestingly, Re-1 alone did show a significant reduction in tau staining versus sham. Overall, the results reflect that pairing BBB opening with a BBB-penetrating therapeutic is worth scaling up with larger cohorts and optimized dosing/schedules.
